Il est le frère de Jonathan LaPaglia, qui a tenu le rôle du chrononaute dans Sept jours pour agir (7 Days). Celui-ci a également joué Brent Langer dans NCIS : Enquêtes spéciales.
Anthony LaPaglia est surtout connu pour le rôle de Jack Malone, qu'il tient depuis 2002 dans la série américaine FBI : portés disparus.
Marié en septembre 1998 à l'actrice australienne Gia Carides, il a une fille Bridget née en janvier 2003.
