Novak Djokovic was the two-time defending champion, but lost in the second round to the 117th ranked Denis Istomin from Uzbekistan. It was the first time since the 2008 Wimbledon Championships, where he lost to Marat Safin, that Djokovic failed to make it past the second round of a major and the first time since the 2006 Australian Open that he failed to make it to the third round in Melbourne. Roger Federer won his fifth Australian Open title, and 18th Major title overall, defeating Rafael Nadal in the final in five sets. With the win, Federer became the first male player to win at least five titles at three Grand Slam tournaments (five at the Australian Open, seven at Wimbledon and five at the US Open). This was the first Grand Slam tournament in which Andy Murray started as World No. 1 and top seed. Murray retained the top ranking despite losing to Mischa Zverev in the fourth round, because second seeded Djokovic needed to win the tournament to regain the No. 1 ranking, but lost in the second round. This was the first time since the 2004 French Open that the top two men's seeds both failed to make it past the fourth round of a major. This was also the first Grand Slam tournament since the 2002 US Open in which Federer was ranked outside the top-10. He went on to reach and win the final of the tournament, making him the oldest man to reach a Grand Slam final since Ken Rosewall made the 1974 US Open final at the age of 39, and the oldest Slam champion since Ken Rosewall had won the 1972 Australian Open at 37 years and 63 days old.

Angelique Kerber was the defending champion, but lost in the fourth round to Coco Vandeweghe. Serena Williams won the title, her 7th Australian title overall and first Australian Open where she did not lose a set during the tournament. She also regained the No. 1 ranking, defeating her sister Venus Williams in the final, 6–4, 6–4. This marks the first time the Williams sisters met in a Grand Slam final since the 2009 Wimbledon Championships and the first time they met in the Australian Open final since 2003. Serena's victory was her 23rd Grand Slam title in singles, surpassing Steffi Graf as the all-time leader in Grand Slam titles in the Open era.

Novak Djokovic was the defending champion, but lost to Dominic Thiem in the quarterfinals. Rafael Nadal won his 10th French Open title and 15th Grand Slam singles title, defeating Stan Wawrinka in the final, 6–2, 6–3, 6–1. Nadal is the only man ever to win 10 singles titles at the same Grand Slam event. He also won this event without losing a set for the third time, thereby tying Björn Borg for the overall men's Grand Slam record.

Garbiñe Muguruza was the defending champion, but lost to Kristina Mladenovic in the fourth round. Muguruza's defeat—along with those of Svetlana Kuznetsova, Venus Williams and Samantha Stosur in the same round—ensured that for the first time in a Grand Slam tournament since the 1979 Australian Open, none of the quarterfinalists were former Grand Slam champions. Jeļena Ostapenko won the title, defeating Simona Halep in the final, 4–6, 6–4, 6–3. This was Ostapenko's first WTA Tour-level singles title. She became the first Latvian player of either sex to win a Grand Slam singles tournament, the youngest woman to win the French Open since 1997, and the first woman since Barbara Jordan at the aforementioned 1979 Australian Open to win a Grand Slam event as her first tour-level singles title. Ostapenko was the first unseeded woman to win the French Open since 1933, as well as the lowest-ranked (47) since computer rankings began in 1975. As a result of Halep's loss in the final, Angelique Kerber retained the WTA no. 1 singles ranking even though she lost in the first round. Kerber's loss marked the first time the top women's seed had lost her first round match at the French Open in the Open era, and the first time this had happened at any Grand Slam tournament since the 2001 Wimbledon Championships. This was the first Grand Slam tournament to feature neither Serena Williams nor Maria Sharapova since the 2002 Australian Open.

Andy Murray was the defending champion, but lost to Sam Querrey in the quarterfinals. Despite his loss, Murray retained the ATP No. 1 singles ranking at the end of the tournament, as Rafael Nadal, Stan Wawrinka, and Novak Djokovic all failed to gain enough ranking points to surpass him. Roger Federer achieved his record eighth Wimbledon and 19th Grand Slam men's singles title, defeating Marin Čilić in the final, 6–3, 6–1, 6–4. Federer thus became the only male player to win the Wimbledon singles title eight times, as well as only the second man in the Open era, after Björn Borg in 1976, to win Wimbledon without losing a set. This was Federer's 70th appearance at a Grand Slam, tying the record for male players and a record-breaking 11th men's singles final at the same Grand Slam tournament. In addition, by virtue of his third-round win over Mischa Zverev, Federer won his 317th Grand Slam singles match, surpassing Serena Williams' record of 316 match wins and giving him the all-time record for the most Grand Slam singles wins by any player, male or female. The tournament marked the fifth time that Nadal and Federer won the French Open and Wimbledon respectively in the same year. This was also the first Grand Slam tournament since the 2009 French Open in which Murray, Nadal and Djokovic all failed to reach the semifinals, and the first time since 2004, that no player will be able to reach all 4 quarterfinals in a year. With his win over Murray, Querrey became the first American man to reach a Grand Slam semifinal since Andy Roddick at the 2009 Wimbledon Championships. For the first time since the 2005 Australian Open, David Ferrer was not seeded at a Grand Slam and the first time since the 2004 US Open that fellow Spaniard Guillermo García López did not play at a Grand Slam, ending his streak of 50 consecutive Grand Slam appearances.

Serena Williams was the two-time defending champion, but did not defend her title because of pregnancy. Garbiñe Muguruza won her second Grand Slam singles title, defeating Venus Williams in the final, 7–5, 6–0. Muguruza became the second Spanish woman to win Wimbledon after Conchita Martínez in 1994. Muguruza also became the first player to defeat both Williams sisters in Grand Slams singles finals. Williams was the oldest player to reach the final since Martina Navratilova, also in 1994, and played her 100th singles match at Wimbledon when she defeated Jeļena Ostapenko in the quarterfinals. Johanna Konta became the first British woman to reach the semifinals since Virginia Wade in 1978, and Magdaléna Rybáriková became the first Slovak woman to reach the semifinals at Wimbledon. Despite losing in the second round, Karolína Plíšková attained the WTA No. 1 singles ranking at the end of the tournament, after Angelique Kerber and Simona Halep, the other contenders for the top ranking, lost in the fourth round and quarterfinals, respectively.

Stan Wawrinka was the defending champion, but was unable to defend the title due to a knee injury that ended his season prematurely. Wawrinka's withdrawal ended his streak of 50 consecutive grand slam appearances, dating back to the 2005 French Open. Rafael Nadal won his 16th Grand Slam title and 3rd US Open title, defeating first time Grand Slam finalist Kevin Anderson, 6–3, 6–3, 6–4. Nadal retained the ATP No. 1 singles ranking after Roger Federer lost in the quarterfinals. It was also Nadal's first hard court title since January 2014. Anderson was the first South African man to reach the US Open singles final since 1965 and any Grand Slam singles final since the 1984 Australian Open, as well as the lowest-ranked male player to reach the US Open singles final since the ATP rankings began in 1973. Anderson was also the tallest Grand Slam finalist in history. Novak Djokovic withdrew due to an elbow injury that ended his tennis season, ending his streak of 51 consecutive grand slam appearances, dating back to the 2005 Australian Open. Accordingly, this was the first men's singles draw at the US Open since 1971 to include neither finalist from the previous year's tournament. The withdrawals of Wawrinka and Djokovic, along with those of Andy Murray, Kei Nishikori (also ending his season), and Milos Raonic, meant that only 6 of the top 11 ranked players competed in the tournament. The draw was retrospectively adjusted after Murray's late withdrawal, but the timing of the withdrawal meant that Nadal and Federer remained in the same half of the draw. Federer broke the men's singles record for total appearances at Grand Slams with his 71st participation. Also, Andrey Rublev became the youngest man to reach the US Open quarterfinals since Andy Roddick in 2001, and Denis Shapovalov the youngest man to reach the fourth round of the US Open since Michael Chang in 1989.

Angelique Kerber was the defending champion, but lost in the first round to Naomi Osaka. This loss would cause Kerber to fall outside the top ten in the WTA rankings for the first time since 2015. Sloane Stephens won her first Grand Slam title, defeating Madison Keys in the final, 6–3, 6–0. It was the first all-American women's final at the US Open since 2002, and the second time in three years that the final featured two first-time Grand Slam singles finalists from the same country. Stephens became the second unseeded woman in the Open era to win the US Open after Kim Clijsters in 2009. For the first time since 1981 at the US Open—and at any Grand Slam event since the 1985 Wimbledon Championships—all four semifinalists were American. Both occasions were also the last time that four players from the same country accomplished these feats. In addition, Kaia Kanepi became the first qualifier to reach the US Open quarterfinals since Barbara Gerken, also in 1981. This was Maria Sharapova's first Grand Slam appearance since the 2016 Australian Open, following a suspension for violating the anti-doping rules. Sharapova was awarded a wildcard into the main draw and advanced to the fourth round, where she lost to Anastasija Sevastova. Garbiñe Muguruza became the new WTA No. 1 player at the end of the tournament despite losing in the fourth round, after Karolína Plíšková (who held the top ranking at the start of the tournament) failed to defend her finalist points from 2016. Eight of the top nine seeds (with the exception of Kerber) were in contention for the no. 1 ranking at the start of the tournament.
