La méthylglyoxal synthase est une lyase qui catalyse la réaction :

Dihydroxyacétone phosphate  $\rightleftharpoons$  méthylglyoxal + P_i.

Cette enzyme intervient dans la voie du méthylglyoxal (en), présente chez certains procaryotes.
